For acute infectious and chronic inflammatory diseases as well as lung cancer, human lung organoids have opened possibilities for precise in vitro research and a deeper understanding of mechanisms underlying lung injury and regeneration.

Organoids are three-dimensional self-organizing structures formed by adult tissue stem cells or pluripotent stem cells. They recapitulate cell-cell, cell-niche interactions in tissue development, homeostasis, regeneration and disease, and provide an in vitro model for drug screening.

In this review, we shall discuss the main type of lung organoids and how to obtain them. We shall also focus on in vivo lung organoid transplantation studies to analyze the viability, engraftment, and maturation of organoids, as well as the effectiveness of the treatment of pulmonary diseases.
